deGrad(GFP-3G86.32) is an engineered F-box protein that targets GFP-tagged proteins for degradation via the ubiquitin pathway. deGrad(GFP-3G86.32) contains the N-terminal 198 amino acids of D. melanogaster slmb (this includes the F-box domain), fused to DARPin(3G86.32), a designed ankyrin repeat protein (DARPin) that specifically binds GFP and a number of its closely related artificial derivatives. Upon expression of deGrad(GFP-3G86.32), target proteins tagged with GFP (or any the other GFP derivatives bound by DARPin(3G86.32)) are specifically polyubiquitinated and rapidly degraded by the proteasome (FBrf0227045).
