Flamindo2 is a genetically encoded fluorescent cAMP sensor. It consists of the Citrine fluorescent protein, into which the cAMP-binding domain from the Mus musculus Rapgef3 gene (MGI:MGI:2441741), flanked on each side by linker sequence, has been inserted (between residues 144 and 146). Flamindo2 was generated by mutagenesis of Flamindo, by optimization of the linker peptides, and has a 2-fold expanded dynamic range and 8-fold increased brightness compared to Flamindo. Flamindo2 shows an excitation peak of 504nm and an emission peak of 523nm. The fluorescence intensity of Flamindo2 decreases by 75% in the presence of a saturating dose of cAMP (PMID:24959857).
