Description

FlyBase curator comment: Tag:AsLOV2(BLITz-Unk) represents an engineered light-regulated TEVp protease target site that corresponds to one of the 'BLITz' target sites described in PMID:28369042, but the particular variant used is unknown or not specified. Tag:AsLOV2(BLITz-Unk) consists of the LOV2 flavin binding domain (including the carboxy-terminal Jα helix) of the Avena sativa Phototropin 1 (PT1) gene, in which the C-terminal end of the Jα helix has been replaced by a cleavage site for the TEVp protease. In the dark, the closed conformation of the LOV2 domain results in the protease target site being inaccessible to TEVp. Upon illumination with blue light, the α-helix unfolds, exposing the protease target site sequence and allowing cleavage by TEVp.
