Description

H{Lw2} is an enhancer trap construct that encodes a lacZ reporter. The enhancer trap sequence is at the 3' end of the construct and consists of weak promoter sequence from the Hsp70 gene (-50 to +271, includes the TATA box but lacks heat shock-inducible sequences, also encodes the first 7 amino acids of Hsp70) fused in-frame upstream of the lacZ coding sequence.

report comment: The enhancer trap allele, lacZHZ50a, is theoretically identical to the allele found in pP{HZ50PL}, however only the 5\' end is actually derived from pP{HZ50PL}; the 3\' end is derived from pP{FZ}.

mapping_comment: Although segment 11, Hsp70Bb::lacZ_4.1 , is the same as that found in pP{HZ50PL}, only the 5\' end (adjacent to hobo 3\' end) is actually derived from pP{HZ50PL}. The 3\' end (adjacent to neoR) is derived from pP{FZ}; fusion at Bsu36I site at 5\' end of lacZ segment.

construct_comment: Useful sites for plasmid rescue (Brian Calvi, personal communication to FlyBase): cloning to left, NdeI, BamHI, MluI; cloning to right, ApaI, NsiI, HindIII. Resulting clones are kanamycin resistant.
