P{FRT(iwoff)} contains a non-functional w minigene consisting of a 5' portion of w, an FRT site, the 3' portion of w (in reverse orientation to the 5' portion) and a second FRT site (in the opposite orientation to the first one). The P{FRT(iwoff)} transgene is generated in vivo by FLP-mediated inversion of the sequence flanked by FRT sites in P{FRT(iwon)}. This process is reversible; FLP can be used to reinvert the FRT flanked sequence in P{FRT(iwoff)} to regenerate P{FRT(iwon)} (which contains the functional w+mW.FRT.hs marker).
