Two tandem copies of a mutated 260bp twi proximal element (PE) (located between -440 and -180 relative to the twi promoter), in which the core sequences of the low affinity TD4 and TD5 dl binding sites in both copies of the PE have been mutated to the optimal sequence to create high affinity sites, followed by twi sequence from -180 to +160bp relative to the transcription start site, are fused upstream of lacZ.
The domain of expression is as forEcollacZtwi.PEe, but the level of expression is higher.
construct_comment: One of a series of constructs characterizing 5\' regulatory regions of twi, in particular the dl binding sites. Additional related constructs listed under P{twi-lacZ.P}.
