Transgenic construct used to study the impact of local chromatin modification on DNA amplification at the origin of replication of the third chromosome chorion gene cluster. Contains: a w+mC marker - five copies of the UAS binding site - a 3.8kb genomic fragment that corresponds to the minimal functional origin sequence of the chorion gene cluster (this fragment includes the ACE3 amplification control element, the Cp18 transcription unit, the Ori-β origin of replication and the Cp15 transcription unit). A chromatin-modifying protein of interest can be tethered to the UAS sites in the P{TT1} transgene by fusing the protein to GAL4(DBD), allowing the impact of chromatin modification on the activity of the origin sequence to be studied.
