Description

The P{PBac.PBss} element acts as a donor element for a gene trap construct. It contains a w+mC marker and a piggyBac based element (PBac{PBss}) which can be mobilized using Tni\piggyBac\T. The piggyBac-based element contains a gene trap cassette that consists of a consensus splice acceptor site, a promoterless EGFP coding sequence tagged at the N-terminal end with Tag:HA (an ATG initiation codon is present in the first translated frame) and an α-tubulin 3' untranslated region for transcription termination. Insertions of the starter P{PBac.PBss} element were not found to express EGFP, but if the PBac{PBss} gene trap element is released using Tni\piggyBac\T, insertions that express EGFP are observed.
