Description

6.45kb XhoI-ApaI genomic fragment that encompasses the osk locus. The two osk translation initiation codons have been mutated (M1L and M139L). In addition, a sequence predicted to adopt a stable hairpin structure has been inserted between the two mutated initiation codons and a frame-shift (CCACTGG instead of CCACCTGG) has been introduced downstream of the second initiation codon. The hairpin structure has been introduced to prevent translation from in-frame methionine codons downstream.
