The P{FRT(Dpy-30L1.YFP)} construct is a targeting construct for disruption of the Dpy-30L1 gene by homologous recombination and can also be used to study the expression of YFP under the influence of the Dpy-30L1 regulatory sequences in the construct.
Contains a DNA segment from the Dpy-30L1 locus, with 4.5kb of upstream and 2.5kb of downstream sequences (relative to the Dpy-30L1 transcription unit) that also includes another four genes (CG6443, CG17118, EMC3 and Nup107). The coding sequence of YFP with its genuine stop codon followed by the SV40 polyadenylation/transcription termination sequence has been inserted in frame immediately following the ATG start codon of Dpy-30L1. The insertion of YFP results in a disruption of the Dpy-30L1 reading frame as well as a deletion of 17 amino acids from the coding region, the truncated Dpy-30L1 sequence is out of frame relative to the ATG-YFP sequence.
