Donor construct for ends-out targeted replacement of the Obp57d and Obp57e loci. The construct contains flanking sequence from 5' of the Obp57d locus (including the CG13430 and Cpr57A genes), followed by a loxP cassette containing a w+* marker, followed by flanking sequence from 3' of the Obp57e locus (including the CG30148 gene). A RS(I-SceI) restriction site is present upstream of the 5' flanking sequence. The entire targeting region is contained within an FRT cassette. The construct is designed to precisely remove both the Obp57d and Obp57e genes, but to leave intact a putative poly-A additional signal (ensuring the termination of any residual transcription that might otherwise affect the expression of downstream genes). The loxP cassette containing the w+* marker can also be removed, leaving only 34bp of the loxP sequence.
