PBac{806.LOX-SVS-2} is a piggyBac-based protein trapping construct. The protein trap sequence is composed of a splice acceptor site, a multiple tag cassette (Tag:StrepII-Venus-Tag:StrepII) in splice frame 2 and a splice donor site. A lox2272 site and a loxP site are present, such that the internal sequence of the construct (including the w^+mC marker and protein trap exon) can be exchanged using recombination-mediated cassette exchange (RMCE). The protein trap cassette is in reverse orientation relative to the PBac ends, so that when the protein trap cassette is in the forward orientation, the components of the transgene are ordered as follows: 3' PBac terminal repeat - lox2272 - w^+mC marker - protein trap cassette - loxP - 5' PBac terminal repeat.
