UAS regulatory sequences drive expression of a transcription unit that expresses two artificial miRNAs that target the w gene (miW-1 and miW-2). These miRNA sequences are embedded in an intron which is followed by an exon containing the coding sequence for GFP.
M{UAS-automiW} acts as a biosensor for AGO2-mediated silencing. The miRNA sequences encoded by the wScer\UAS.automiW transcription unit in the construct are complementary to the w+mC marker present in the construct, thus if AGO2-mediated silencing is functional, expression of wScer\UAS.automiW in a w[-] background using a GAL4 driver results in flies with white eyes. If AGO2-mediated silencing is compromised, this silencing is inhibited, resulting in flies with pigmented eyes.
