Ubi-p63E regulatory sequences drive expression of a fusion protein consisting of Mmus\Cd8a and the GAL4 coding region, separated by sequence derived from Diap1. The Diap1 sequence consists of the caspase-3-binding and cleavage domain (amino acid residues 2-147) in which the DQVD caspase cleavage site has been changed to DQVA (residues residues 21 and 22 have also been mutated from NN to GV). This construct serves as a caspase-insensitive control in the CasExpress system, as the mutated DQVA site can no longer be cleaved by caspase: the Mmus\Cd8a sequence tethers GAL4 to the plasma membrane, rendering it inactive.
