The PBac{EGFP-IV} element contains an artificial exon composed of EGFP (with its own translation start site) flanked by splice acceptor and splice donor sites. This exon is flanked by minimal piggyBac terminal sequences, and thus the total length of the element is only 962bp. The element is designed to act as a protein trap element when inserted into a coding intron of an endogenous gene in the correct orientation. The presence of the translation start site means that insertions into the 5'UTR of an endogenous gene may act as a promoter trap, also resulting in EGFP expression.
