Description

The Ubi-p63E promoter drives expression of a fusion protein comprised of a transmembrane domain (Tag:M(mCd8a)), amino acids 2-147 of Diap1, GAL4 and a Tag:MYC tag. The caspase cleavage site in the Diap1 sequence has been mutated to DQVAGV, so that it can no longer be cleaved. The Diap1 sequence also includes the a D135R mutation. The transmembrane domain tethers the fusion protein to the membrane regardless of caspase activity, preventing the GAL4 sequence from entering the nucleus, and thus it is unable to drive expression of UAS-regulated responder transgenes. This transgene is intended as a control for the Caspase-sensitive P{Ubi-p63E-mCD8::DQVD::GAL4} sensor.
