Description

'Zld3'(a 56bp neutral DNA sequence into which three consensus zld binding sites have been embedded), the 'Bcd3' enhancer (a synthetic enhancer containing three binding sites for bcd), a 73bp linker and the Drosophila synthetic core promoter (DSCP) are fused upstream of mNeonGreen that is tagged with three nuclear localization signals; one (Tag:NLS(sd)) at the N-terminal end and two (Tag:NLS(SV40-largeT), Tag:NLS(Unk)) at the C-terminal end of the fluorescent reporter. The coding sequence has been codon optimized for expression in D. melanogaster. 5'UTR sequence (IVS+syn21) containing a translational enhancer and 3'UTR sequence (p10) is also present.
