Description

The M{FlyORF-TaDa} transgene is designed to allow in vivo conversion of a compatible FlyORF line into a line that can be used for Targeted DamID (TaDa). It contains a donor cassette in which UASt regulatory sequences are fused upstream of a bicistronic transcript (represented in FlyBase by Ecol\damUAS.FlyORF-TaDa) that encodes two open reading frames separated by stop codons (the upstream ORF encodes myristylated GFP and is simply present to reduce translation of the Tag:Dam-containing downstream ORF). The downstream ORF comprises Tag:Dam, a Tag:MYC epitope and an in-frame FRT5 site followed by a stop codon. In the absence of FLP, this donor cassette can act as a Tag:Dam-only control for DamID signal normalization. In the presence of FLP recombinase, the donor cassette can be 'swapped' in to a compatible FlyORF insertion line that contains an ORF of interest and which is in the same genomic insertion site on the homologous chromosome. This results in a converted line in which UASt regulatory sequences drive expression a Targeted DamID (TaDa) line of the ORF of interest, with the donor cassette inserted upstream of the ORF of interest such that the Tag:Dam-containing ORF of the donor cassette is in-frame with the ORF of interest. FlyORF insertion lines that contain an FRT5 site in between their promoter and ORF are compatible with a M{FlyORF-TaDa} insertion at the same genomic insertion site.
