Transgenic construct used to test the ability of a kel fragment to mediate stop codon readthrough. UAS regulatory sequences drive expression of a reporter in which two fluorescent proteins (mCherry and GFP) are separated by a kel gene test fragment (that extends from 84 nucleotides 5' to 321 nucleotides 3' relative to the UGA kel ORF1 stop codon) which is flanked on each side by a T2A sequence. Readthrough efficiency can be calculated from the ratio of GFP produced by this transgenic construct compared to a matched control in which the UGA stop codon of the kel fragment has been mutated to an in-frame UGG codon (P{UAS-mCh-T2A-UGG-321-T2A-GFP}).
