Description

Transgene containing one complete rDNA unit comprising the non-transcribed spacer (NTS), which includes elements that regulate RNA polymerase I (Pol I) transcription, and the complete transcribed portion that generates the 47S pre-rRNA. The pre-rRNA contains a 5' external transcribed sequence (5' ETS) and internal transcribed spacers (ITS), which are removed from the pre-rRNA during its processing into mature 18S, 5.8S, 2S and 28S rRNAs. The transgene also has 28 bp of the 28S rDNA from the upstream unit. The transgene is marked by insertion of a 21 bp unique identification sequence (UID) into the ETS. In addition, the sequence of the R1A1-element transposon is inserted into its natural integration site within the 28S rRNA.
