10xUAS regulatory sequences drive expression of a dicistronic transcript that encodes two open reading frames separated by a 2A linker (this results in separate translation of each protein). The first ORF encodes 'post-mGRASP' and the second ORF encodes dTomato. post-mGRASP (PMID:22138823) is composed of an N-terminal signal peptide, the spGFP1-10 split GFP fluorescent protein fragment, and portion of the mouse mNlgn1 gene that targets the protein to the postsynaptic membrane (Tag:TS(mNlgn1)).
