UASt sequences regulate expression of a mutated form of Rbf that lacks the IE domain (residues 728-786 are deleted). The coding sequence is tagged at the N-terminal end with both 2xTag:FLAG and dCas9(D10A.H840A) and at the C-terminal end with 2xTag:FLAG. The presence of the dCas9(D10A.H840A) sequence means that the Rbf protein can be recruited to specific promoters using appropriate gene-specific guide RNAs.
