10xUAS sequences regulate expression of a "sponge" construct designed to knockdown the miR-34-3p, miR-11-3p, miR-92a-3p, miR-92b-3p, miR-8-3p and miR-315-5p miRNAs. The sponge sequence consists of repeats of a 21bp sequence that is complementary to the targeted miRNA except for mismatches at positions 9-12, with a 4bp linker between each repeat; the number of repeats for each miRNA are: miR-34-3p (20), miR-11-3p (20), miR-92a-3p (10), miR-92b-3p (10), miR-8-3p (20) and miR-315-5p (20). The sponge sequences are inserted into 3'UTR sequence downstream of the DsRed2 coding region.
