Description

Imprecise excision of the progenitor insertion, resulting in a 2098bp deletion that extends 970bp upstream and 1128bp downstream of the original insertion site. This deletion removes the first exon of the Ptp61F m and n isoforms.

Comment:

2098 bp deletion resulting from the imprecise excision of P{RS5}Ptp61F5-HA-2445, which removes sequence extending from 970 bp upstream to 1128 bp downstream of the insertion site.

Detailed Description

Homozygous flies are normal in size (as measured by wing centroid size).

Homozygous females show a significant reduction in life span. Male fertility is normal, but female fecundity (number of eggs laid per fly per day) is reduced compared to wild type. The number and morphology of somatic cells surrounding the developing egg chambers appear normal in mutant ovaries. However, late stage egg chambers (particularly stage 10 or later) are missing, and there is an accumulation of germline stem cells and/or early stage egg chambers. There is an approximately 8-fold increase in the number of apoptotic pre-stage 10 egg chambers compared to wild type.

Expression of Ptp61FScer\UAS.n.T:Hsap\MYC under the control of Scer\GAL4arm.PU fails to rescue the reduced lifespan of Ptp61FΔ flies. The reduced fecundity of Ptp61FΔ females is partially rescued.

Simultaneous co-expression of Ptp61FScer\UAS.m.T:Hsap\MYC and Ptp61FScer\UAS.n.T:Hsap\MYC under the control of Scer\GAL4arm.PU rescues the reduced lifespan of Ptp61FΔ flies. The reduced fecundity of Ptp61FΔ females is also largely rescued.

Expression of Ptp61FScer\UAS.m.T:Hsap\MYC under the control of Scer\GAL4Act.PU fails to rescue the reduced lifespan of Ptp61FΔ flies.

Expression of Ptp61FScer\UAS.m.T:Hsap\MYC under the control of Scer\GAL4arm.PU partially rescues the reduced fecundity of Ptp61FΔ females.
