Construct: bcd 5' sequences are exchanged with Xenopus β-globin 5' nontranslated sequences.
In vitro transcribed bcdTN3 mRNA can induce anterior development at any ectopic position along the anterior-posterior axis. Injection into the posterior pole of bcd6 embryos suppresses posterior development and induces the formation of head and thoracic structures at the posterior end. Injection into the middle of the embryo induces gnathal and thoracic structures at the site of injection.
Carried in plasmid "bcdTN3" and in vitro transcribed in wheat germ and reticulocyte lysate systems for injection into bcd6 females to study rescue of the mutant phenotype.
Carried in plasmid "BcdTN3", protein expressed from the Sp6 promoter, radioactively labeled in a reticulocyte translation system and interaction with various bcd domains is assayed using coimmunoprecipitation analysis.
