UASt regulatory sequences drive expression of mature SP (lacking the signal sequence) that is tagged at the N-terminal end with Tag:M(hGGT1). This results in a membrane-bound protein: the SP portion is expressed extracellularly, but remains attached to the expressing cell (via the membrane located Tag:M(hGGT1) tag).
Expression of SPScer\UAS.T:Hsap\GGT1 in virgin females using Scer\GAL4ppk.PG, Scer\GAL4fru.P1.D or Scer\GAL4dsx.KI reduces receptivity (readiness to mate) compared with wild-type and increases oviposition similar to mating.
Virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4dsx.KI show levels of sexual receptivity comparable to mated control females, which are significantly lower than the receptivity of virgin control females. They also show significant rejection behaviours toward courting males, as demonstrated by an increased frequency of ovipositor extrusions, and show an increase in oviposition, similar to the levels seen in mated control females. Compared to control virgin females, the mutant virgin females show a marked decrease in locomotion while being courted and also show reduced levels of activity while the male is not courting.
Wild-type naive males court virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4dsx.KI significantly less than control virgin females, but this reduction in courtship index is not seen in experiments with decapitated mutant virgin females.
The reduction in sexual receptivity and increase in oviposition seen in virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4dsx.KI compared to control virgin females is partially rescued if the flies also carry Scer\GAL80ppk.PY.
Virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4Cha.7.4 show reduced sexual receptivity compared to control virgin females.
Virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4fru.P1.D show a strong decrease in sexual receptivity accompanied by rejection behaviours toward courting males (including ovipositor extrusion) and an increase in egg laying to a comparable extent to mated wild-type females.
Virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4ppk.PG show a decrease in sexual receptivity accompanied by rejection behaviours toward courting males and an increase in egg laying to a comparable extent to mated wild-type females.
Virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of either Scer\GAL4Ilp7.PY or Scer\GAL4Or67d-GAL4-1 show normal postmating behaviours.
The reduction in sexual receptivity and increase in oviposition seen in virgin females expressing Acp70AScer\UAS.T:Hsap\GGT1 under the control of Scer\GAL4fru.P1.D compared to control virgin females is largely rescued if the flies also carry Scer\GAL80ppk.PY.
Scer\GAL4C357, Scer\GAL455B, Scer\GAL4C420, Scer\GAL4C370 and Scer\GAL4C502b-mediated expression causes only a slight drop in female receptivity whereas ovulation is very low. Scer\GAL429BD, Scer\GAL4156Y, Scer\GAL419Y, Scer\GAL49Y and Scer\GAL47B-mediated expression allows no female receptivity but stimulated very high levels of ovulation.
