Mtork03905 mutant MARCM clones in central brain neuroblasts show reactivation defects as high proportion of the cells retain cellular process and vast majority are not mitotically active at 96h after larval hatching.

In adult testes, Mtork03905-homozygous germline stem cell clones are fewer in number than control germline stem cell clones at day 1 after clone induction, becoming negligible or inexistent at days 2 or 7 after clone induction, respectively; at day 1 after clone induction, mosaic testes bearing Mtork03905 mutant clones in an otherwise Mtork03905 heterozugous background display increased apoptosis as compared to controls, which is limited to outside of the germline stem cell region.

Somatic cyst stem cells are much fewer in number in Mtork03905-homozygous clones within mosaic testes, being negligible at days 1 and 2 post clone induction and inexistent at day 4 post clone induction, as compared to control clones within control testes; no cell death is detected in the somatic cyst stem cell lineage of Mtork03905-homozygous clones.

Among 300 embryos, only one homozygous Mtork03905 first instar larvae emerged.

Expressing either mad2Scer\UAS.cLa or Mps1Scer\UAS.cLa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 suppresses the germline stem cell loss present in homozygous Mtork03905 germline stem cell clones in adult testes, 2 days post clone induction.

Expression of Apc2Scer\UAS.cUa under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 partially rescues the decreased number of germline stem cells in homozygous Mtork03905 within mosaic testes, as compared to control clones in control testes.
