Description

The P{lacW} insertion is 71bp upstream of the ATG start of Cas.

The P{lacW}Cask03902 insertion maps within both Cas and mdy (nested genes on opposite strands).

Surviving escapers expressing Kap-α1Scer\UAS.cMa under the control of Scer\GAL4e22c in a Cask03902/+ background have tergites that appear thinner than normal and retain only the posterior pigmented band, resulting in an apparent expansion of the intertergal region.

Comments

The lethality caused by the P{lacW}Cask03902 insertion is due to its effect on Cas and not mdy; the lethality is rescued by Cas+tTa, which contains the entire Cas gene but lacks the mdy transcription start site and sequences encoding the N-terminal half of the mdy protein.

Comments

After six generations of outcrossing the lethality continued to segragate with the P{lacW} insertion.

Precise excision of the P{lacW} element reverts the recessive lethal phenotype. Phenotypic analysis provides an allelic series: Cask03902/Df(2L)H20 > CasEMS25 > Casts28/Df(2L)H20 > CasEMS39.
