Full length C3G tagged with 6 copies of the Tag:MYC tag is expressed under the control of UASt regulatory sequences. The C3G sequences are also tagged at the 5' end with Tag:M(c-SRC), to produce a membrane-targetted protein.

C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 driven by Scer\GAL4Mef2.PR at 18C allows development of larvae. The larvae display severe morphological muscle defects, disruption of actin organisation in somatic muscles and detachment of the ventral longitudinal muscles compared to controls.
Flies expressing C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 under the control of Scer\GAL4GMR.PF have a rough eye phenotype with varying degrees of severity depending on the P{UAS-C3G.T:Myc.T:Myr3} insertion line used. Flies with an intermediate phenotype have a moderately rough eye surface with differentiated lenses and bristles. All ommatidia contain supernumerary photoreceptor cells, with up to 5 or 6 additional cells in some cases. Most of these extra photoreceptor cells have small rhabdomeres and are located in the distal portion of the retina, suggesting that they are R7 cells. Some pigment cells are present, but they fail to form a lattice surrounding the ommatidia. Flies with a strong phenotype have reduced eyes and the surface appears rough and folded. There are very few bristles on the surface of the eye and lens facets are not discernible. The ommatidial lattice is severely disrupted; fewer clusters of photoreceptor cells are seen than normal, the number of cells per cluster and the arrangement of the cells is irregular and the rhabdomeres often do not span the entire thickness of the retina and have an elongated morphology. Wing discs expressing C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 under the control of Scer\GAL4dpp.blk1 show an increase in BrdU incorporation in a band corresponding to the domain of Scer\GAL4dpp.blk1 expression. Adult wings of flies expressing C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 under the control of Scer\GAL4dpp.blk1 show a massive broadening of the third vein.

Sose4G or drke0A has no effect on the eye phenotype caused by C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 expressed under the control of Scer\GAL4GMR.PF. The wing vein phenotype seen in flies expressing C3GScer\UAS.T:Hsap\MYC,T:Ggal\Myr3 under the control of Scer\GAL4dpp.blk1 phenotype is dominantly suppressed by Ras85De1B, ksrS-638, rlx-3-30 or Df(3L)R-G7, and is unaffected by Sose4G or drke0A.
