Syx1AΔ229 and Syx1A06737 flies rescued with Syx1A+t13.5, like wild-type, undergo peristaltic muscle contractions prior to hatching. They also show a robust response to tactile stimulation. Syx1AΔ229 embryos rescued with Syx1AI216A exhibit wild-type cuticle phenotypes. Using whole-cell patch-clamp analysis, Syx1AI216A in a Syx1AΔ229 null background shows a very significantly enhanced evoked junctional current amplitude compared to Syx1A+t13.5 controls (over 150% of wild-type amplitude). There is no significant change in the amplitude of spontaneous mEJCs.
