Overexpression of howL.Scer\UAS in the germline via Scer\GAL4nos.UTR.T:Hsim\VP16 results in testes larger in size than in wild-type, with the most notable difference being an expansion of the apex of the testis. Overexpression of howL.Scer\UAS causes a hyper-proliferation of spermatogonial cells.
In wild-type testes, the maximum number of spermatogonia within a cyst is 16, which differentiate into 16 spermatocytes. In Scer\GAL4nos.UTR.T:Hsim\VP16->howL.Scer\UAS testes, cysts of 32 and 64 spermatocytes are frequently observed. This failure to exit mitotic division is not permanent, as germ cell tumours are not apparent, and spermatogonial cysts with increased numbers can differentiate into spermatocytes. Cysts containing 32 spermatocytes are able to undergo meiosis correctly; cysts of 128 early spermatids are observed, which is double the number found in wild-type.
Expression of howL.Scer\UAS under the control of Scer\GAL4nos.PG has little effect on germline stem cell development.

Expression of howL.Scer\UAS in early germline stem cells under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 rescues the howr17/howr4 germline stem cell phenotype.
