A Stop codon is inserted 6 amino acids after the transmembrane domain (replacing residue Y276). The last amino acid before the stop is also changed (amino acid replacement I275R). The truncated cDNA is driven by a 5kb grk genomic fragment.
The addition of one copy of grkΔC into a homozygous grk2B6 background produces mothers whose eggs are rescued almost to wild-type (both anterior posterior and dorsal ventral defects are rescued). All embryos have aeropyles, and less than 1/4 of the sets of dorsal appendages are fused. Extra copies led to dorsalisation of the egg.
