An almost full length copy of Su(var)205 (the first 10 N terminus amino acids are missing) are fused in frame to DNA binding domain of Scer\GAL4. This fusion protein is driven by heatshock promoter.
Heat shock induction of one Su(var)205hs.T:Scer\GAL4 transgene results in variegation in P{Winkelried}A7 eyes. Variegation is not seen after Su(var)205hs.T:Scer\GAL4 induction in P{Winkelried(-FRT)}A7 eyes (P{Winkelried(-FRT)}A7 is the same as P{Winkelried}A7 but without Scer\GAL4-binding sites). Furthermore, variegation is not seen in P{Winkelried}A7, P{Winkelried(-FRT)}A7 eyes. Heat-shock induced expression of Su(var)205hs.T:Scer\GAL4, induces loops between the P{Winkelried}A7 insertion site and intercalary, pericentric or telomeric heterochromatin in polytene chromosomes. This effect is not seen with the P{Winkelried}D insertion.
