Imprecise excision of P{lacW}nrv2k04223 has deleted the first two and most of the third common exons of nrv2. The genomic sequence across the deletion breakpoints is AAGGCCCTCGCTATAACCAAGGAGAATAAC.

In nrv2nwu3 homozygous embryos, beginning at the time of tracheal tube expansion (stage 15), multicellular tracheal tubes become increasingly abnormal. By stage 16 the average dorsal trunk length is significantly greater than wild type (P<0.005) (113+/-2% mean+/-s.e.m., n>5, normalized to stage 16 wild-type value) and all tube diameters are irregular with expansions and constrictions along their lengths. Defects are also present in regions of single cell tubes formed by autocellular junctions, particularly near the ends of the ganglionic branches where there are lumenal staining discontinuities. nrv2nwu3/nrv223B embryos from nrv2nwu3 germline clone mothers are indistinguishable from nrv2nwu3 homozygotes with respect to their tracheal patterning. Unlike wild-type, post stage 15 trachea in nrv2nwu3 homozygotes are unable to exclude from their lumens, a fluorescently labelled 10kDa dextran injected into the body cavity. This is consistent with these trachea lacking a functioning septate junction barrier.

sinunwu7 nrv2nwu3 double mutants have cyst-like structures in the tracheal ganglionic branches.

nrv2nwu3; cora5 embryos have the same phenotype as either single mutant. The tracheal phenotypes of nrv2nwu3; GliJ29Δ7b double homozygous embryos are not more severe than that of nrv2nwu3 homozygous embryos. The tracheal phenotypes of nrv2nwu3 homozygotes are exacerbated by convk06507b/convk06507b or mmyk13717b/mmyk13717b.
