Comment:

A 13bp deletion (nucleotides 644-656 of cDNA) leads to a frameshift at Asp203 adding 16 novel amino acids followed by a stop codon. The location of the deletion was mapped relative to the cDNA from GB:AF247501.

Detailed Description
Statement

pieE1-16/pieEB3 mutant adult survivors exhibit a marked rough eye phenotype.

pieE1-16 mutant eye disc clones exhibit increased apoptosis compared with wild type controls, and the resulting eyes contain very few mutant clones.

pieE1-16 mutant male germline stem cell clones are seen at two days after induction but rapidly lost by 5-7 days.

The division rate of pieE1-16 mutant female germline stem cell clones is reduced compared to controls, and the cell cycle is disrupted.

pieE1-16 intestinal stem cell clones in the midgut generate significantly fewer cells compared with wild-type clones.

Somatic clones of pieE1-16 cells in the eye disc have many apoptotic cells (evenly distributed throughout the clone) and small ommatidia with a variable shortfall in numbers of photoreceptors and cone cells. In adult eyes these clones are rough.

Additional Comments
Genetic Interactions
Statement
Reference

One copy of foxo25 significantly suppresses the male germline stem cell (GSC) loss and reduced GSC division rate seen in pieE1-16 mutant clones.

One copy of foxo25 significantly suppresses the increased apoptosis seen in pieE1-16 mutant eye disc clones, and clones are seen in adult eyes.
