Expression of sniScer\UAS.cBa under the control of Scer\GAL4Act5C.PI under normal oxygen conditions does not extend the lifespan of adult flies. Expression of sniScer\UAS.cBa under the control of Scer\GAL4Act5C.PI under hyperoxia (99.5% oxygen) results in a significant extension of the mean and maximum lifespan (43% and 44.4% respectively) compared to control flies. In addition, the flies expressing are substantially protected from the brain degeneration that is normally induced by hyperoxia. Expression of sniScer\UAS.cBa under the control of Scer\GAL4Act5C.PI counteracts the effect of age on walking speed, improving the performance of 20- and 30- day old flies in a negative geotaxis assay by 63% and 78.8% respectively.
sniUAS.cBa, Scer\GAL4elav-C155 is a suppressor of short lived phenotype of Hsap\APPArctic.UAS.Tag:SS(nec), Scer\GAL4elav-C155
sniUAS.cBa, Scer\GAL4elav-C155 is a non-suppressor of abnormal locomotor behavior phenotype of Hsap\APPAβ42.UAS.Tag:SS(nec), Scer\GAL4elav-C155
Overexpression of sniScer\UAS.cBa partially suppresses the decreased survival phenotype of flies expressing Hsap\APPArctic.Scer\UAS.T:nec under the control of Scer\GAL4elav-C155.
Overexpression of sniScer\UAS.cBa does not significantly suppress the decline in locomotor function of flies expressing Hsap\APPScer\UAS.T:nec under the control of Scer\GAL4elav-C155.
The neuronal and glial cell death that is seen in the lamina of sni1 adults as they age is rescued by expression of sniScer\UAS.cBa under the control of Scer\GAL4elav-C155. The reduced lifespan of sni1 flies seen under normal oxygen conditions and under hyperoxia (99.5% oxygen) is rescued by expression of sniScer\UAS.cBa under the control of Scer\GAL4Act5C.PI. The impaired performance of sni1 flies in negative geotaxis experiments is improved by expression of sniScer\UAS.cBa under the control of Scer\GAL4Act5C.PI.
