P{XP}Nrx-1d08766 is located approximately 200bp upstream of Nrx-1.
P{XP}Nrx-1d08766 is inserted 198b upstream of the translation initation codon of the Nrx-1.
Nrx-1d08766 heterozygous third instar larvae do not display any neuromuscular junction (NMJ) morphological defects (the total bouton area, number of boutons as well as number of branch points per NMJ is comparable to wild-type) but the active zone density at the NMJ is significantly increased relative to controls. Larval locomotor speed is not affected compared controls and the average cell body size of cells in the ventral nerve cord is also not significantly different.
Associative learning is reduced in Nrx-1d08766 mutants (testing for their ability to associate odors with a fructose reward in an individual-animal assay).
Hsap\DISC1UAS.Tag:HA, Nrx-1[+]/Nrx-1d08766 has abnormal locomotor behavior | third instar larval stage phenotype
Nrx-1[+]/Nrx-1d08766 is a suppressor of NMJ bouton phenotype of Hsap\DISC1UAS.Tag:HA, Scer\GAL4αTub84B.PL
Hsap\DISC1UAS.Tag:HA, Nrx-1[+]/Nrx-1d08766 has neuromuscular junction | third instar larval stage phenotype
The reduced total neuromuscular junction (NMJ) bouton area in third instar larvae expressing Hsap\DISC1Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4αTub84B.PL is rescued by combination with a single copy of Nrx-1d08766 but this also results in significant decrease in the number of branch points per NMJ, which is not observed in either of the single mutants alone. Number of boutons per NMJ remains comparable to wild-type.
Similarly, Scer\GAL4αTub84B.PL>Hsap\DISC1Scer\UAS.T:Ivir\HA1;Nrx-1d08766/+ larvae show significant decrease in both average and peak locomotor speed, a phenotype not seen in either of the single mutants. Cell body size of neurons in the ventral nerve cord is not affected and comparable to wild-type.
