UASt regulatory sequences drive expression of a mutant form of SERCA that carries the amino acid replacement E442K (the lesion that is present in SERCAKum170).
Expression of Ca-P60AKum170.Scer\UAS under the control of either Scer\GAL4elav-C155, Scer\GAL4Cha.7.4, Scer\GAL4C380, Scer\GAL4arm.PS or Scer\GAL4shi.PS does not result in any observable paralysis in either larvae or adults which have been incubated at the restrictive temperature of 40oC for 3 minutes. Expression of Ca-P60AKum170.Scer\UAS under the control of either Scer\GAL4αTub84B.PL, Scer\GAL4Mhc.PW or Scer\GAL4how-24B results in paralysis in larvae which have been incubated at the restrictive temperature of 40oC for 3 minutes, but does not result in adult paralysis at this temperature. Expression of Ca-P60AKum170.Scer\UAS under the control of Scer\GAL4Mef2.PR results in paralysis in larvae or adults which have been incubated at the restrictive temperature of 40oC for 3 minutes. The dorsal longitudinal muscles (DLMs) of adults expressing Ca-P60AKum170.Scer\UAS under the control of Scer\GAL4Mef2.PR show broadening of spontaneous action potentials with occasional failures at the restrictive temperature. At this temperature, evoked responses also show defects; spike broadening is followed by frequent failures.
