aldGS13084 mutants show segregational defects for achiasmate homologs. The frequency of achiasmate X chromosome nondisjunction is increased more than 15-fold in FM7a/X; aldGS13084 females, compared to controls. The frequency of nondisjunction is even greater (>65-fold higher than controls) in FM7a/X; aldGS13084/Df(3R)ED5780 females and these mutants also show a ~10% frequency of chiasmate X chromosome nondisjunction. Both FM7a/X; aldGS13084 and FM7a/X; aldGS13084/Df(3R)ED5780 mutants show increased levles of achiasmate 4th chromosome nondisjunction. ald1 neuroblasts exhibit a failure of the meiotic spindle checkpoint. Following treatment with colchicine, the checkpoint of wild-type neuroblasts is activated and cell arrest in metaphase. In contrast, almost 35% of aldGS13084 neuroblasts and ~7.5% of aldGS13084/+ neuroblasts fail to arrest, as shown by precocious chromatid separation.
