Comment:

G to A nucleotide change at the second or third position of the Trp codon leads to a nonsense mutation (exact site of mutation unspecified). Site of nucleotide substitution in mutant inferred by FlyBase based on reported amino acid change.

ave108V mutant clones generated in third instar eye discs lack R1 and R6 photoreceptors, have a reduced number of R3 and R4 cells and have very few cone cells. ave108V expression in these clones indicates a partial failure of G1 arrest and there appears to be a higher level of cell death within the clones.

The ave108V mutation affects wing development. Induction of ave108V mutant clones results in a loss of wing veins in the adult wing. Furthermore, the wing discs of homozygous ave108V third instar larvae show greatly reduced notum primordia.

Mutant clones that express Ras85DV12.Scer\UAS, under the control of Scer\GAL4da.G32, in a ave108V background show the ave108V photoreceptor differentiation phenotype. The Scer\GAL4da.G32>Ras85DV12.Scer\UAS ectopic photoreceptor phenotype is therefore suppressed. In contrast, when phlScer\UAS.F179 is expressed under the control of Scer\GAL4da.G32 in ave108V mutant clones, ectopic photoreceptor cells are induced and the ave108V phenotype is suppressed.

Comments

Neither aveCC9-36B nor aveCC9-20A can complement the lethality ave108V.

Expression of aveScer\UAS.T:Ivir\HA1, under the control of Scer\GAL4da.G32, in ave108V mutant clones of the eye disc rescues the photoreceptor differentiation defect.
