1 day memory is impaired after spaced training, but not after massed training in an olfactory associative conditioning assay in flies expressing TequiladsRNA.Scer\UAS under the control of Scer\GAL4G0431. The defect in memory after spaced training is still seen if the temperature sensitive Scer\GAL80ts.αTub84B allele is used to restrict Scer\GAL4 expression to the adult stage.

RNAi-mediated Tequila knockdown, achieved by expression of TequiladsRNA.Scer\UAS under the control of Scer\GAL4Mef2.247, results in a decrease in 24-hour long-term memory. 24-hour memory after massed training is unaffected , as are learning and 2-hour memory after single conditioning. No alteration of mushroom body morphology can be observed. The long-term memory phenotype induced by RNAi knockdown is reversible as flies expressing TequiladsRNA.Scer\UAS driven by Scer\GAL4Mef2.247.Switch show the phenotype while flies are fed with RU486, but show a normal 24-hour long term memory capacity following transferral to food without RU846, thus preventing Scer\GAL4Mef2.247.Switch from driving TequiladsRNA.Scer\UAS expression.
