Expression of vitoGD10890 using Scer\GAL4ey.PU results in a reduced and rough adult eye. In the eye primordium, posterior retinal differentiation still occurs, but a very significant reduction in eye disc size is visible, particularly in the anterior proliferative region.

Scer\GAL4Ubi-mediated expression of vitoGD10890 results in a significant developmental delay.

vitoGD10890-expressing clones induced early in larval development and analysed in third instar eye discs are significantly smaller than control clones, both in the peripodial epithelium and the eye disc proper. This is not due to a reduction in cell size.

Scer\GAL4ey.PU, vitoGD10890 eye discs exhibit significantly more cells undergoing apoptosis compared to controls.

Scer\GAL4ptc-559.1-mediated expression of vitoGD10890 results in reduced larval salivary gland growth. There is a substantial reduction in the packaging of nucleolar components and nucleoli exhibit an overall granular organization. Salivary gland cells are significantly smaller than controls owing to a reduction in cytoplasmic volume; nuclear volumes are not significantly reduced.

Adults expressing vitoGD10890 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

Expression under the control of Scer\GAL4pnr-MD237 results in bristle morphology defects on the notum in 0% or 80-90% of the Scer\GAL4pnr-MD237 expression domain, depending on the insertion line used.

Heterozygosity for Df(3L)H99 in the Scer\GAL4ey.PU, vitoGD10890 background almost completely suppresses the increased apoptosis phenotype, but eye disc size is only partly rescued.

Homozygosity for p535A-1-4, or co-expression of p53R155H.Scer\UAS.Ex, in the Scer\GAL4ey.PU, vitoGD10890 background does not rescue the increased apoptosis phenotype or eye disc size.

Co-expression of bskDN.Scer\UAS in the Scer\GAL4ey.PU, vitoGD10890 background partially suppresses the increased apoptosis phenotype, but eye disc size is not rescued.

Co-expression of vitoGD10890 reduces the size of Scer\GAL4Act5C.PI-indcued dmScer\UAS.cZa clones to control levels. This is the consequence of partial reversion of the dmScer\UAS.cZa-induced cell size increase together with a decrease in the number of cells per clone.

Co-expression of vitoGD10890 considerably attenuates the increased cell size seen in Scer\GAL4ptc-559.1, dmScer\UAS.cZa salivary glands. This is a result of significant reductions in cytoplasmic and nuclear volumes. The nucleoli display a segregated organization, with a peripheral granular region surrounding an internal fibrillar area.

Co-expression of Hsap\NOL12Scer\UAS.T:Avic\GFP in the vitoGD10890, Scer\GAL4ey.PU background fully rescues eye disc growth.

Coexpression of BacA\p35Scer\UAS.cHa in the Scer\GAL4ey.PU, vitoGD10890 background almost completely suppresses the increased apoptosis phenotype, but eye disc size is only partly rescued.

Comments

Co-expression of vitoScer\UAS.cMa in the vitoGD10890, Scer\GAL4ey.PU background significantly rescues eye disc size, adult eye size and morphology, and several other cellular parameters.
