Approximately 40% of the expected staiB200 mutant homozygous progeny from a heterozygous cross survive to the adult stage.

stairdtp/staiB200 mutant larvae exhibit a posterior paralysis "tail flip" phenotype. When they crawl along the surface of a substrate they exhibit a crawling behaviour in which the posterior body segments sharply flip upward after each peristaltic wave of muscle contraction during the crawling cycle.

77% of homozygous staiB200 mutant larvae exhibit a posterior paralysis "tail flip" phenotype. When they crawl along the surface of a substrate they exhibit a crawling behaviour in which the posterior body segments sharply flip upward after each peristaltic wave of muscle contraction during the crawling cycle. Of the mutant exhibiting this paralysis 70% have a robust tail flip while 30% have a mild tail flip. No posterior paralysis is observed in staiB200/+ larvae and they are able to crawl normally.

89% of staiB200/Df(2L)Exel6015 mutant larvae exhibit a posterior paralysis "tail flip" phenotype. When they crawl along the surface of a substrate they exhibit a crawling behaviour in which the posterior body segments sharply flip upward after each peristaltic wave of muscle contraction during the crawling cycle. Of the mutant exhibiting this paralysis 76% have a robust tail flip while 24% have a mild tail flip. No phenotype is observed in staiB200/+ or Df(2L)Exel6015/+ mutant larvae.

staiB200 mutant adults have a reduced lifespan at 25[o]C, living on average 47 days compared to ~78 in wild type.

staiB200/Df(2L)Exel6015 mutant adults have a reduced lifespan at 25[o]C, living on average 35 days compared to ~78 in wild type.

21 day old staiB200 mutant flies exhibit bang-sensitive paralysis. 21% of males exhibit paralysis with a mean recovery time of ~49 seconds, and ranging from 10 seconds to 3 minutes. Upon recovery the flies enter a hyperactive phase, in which they vigorously flap their wings and move their legs for approximately 4-6 seconds This bang-sensitive phenotype is not observed in 1, 7 and 14 day old flies. By 42 days post-eclosion 76% of adults exhibit a bang-sensitive phenotype, with an increase in the mean recovery period to ~77 seconds. By 56 days the phenotype is seen in 92% of flies, with a recovery time of 145.4 seconds. The phenotype is seen in staiB200/+ adults, but with a delayed onset and a marked reduction in the recovery period following paralysis compared to homozygous staiB200. The bang-sensitive phenotype is first observed in staiB200/+ mutant flies at 42 days, with 25% of flies exhibiting the phenotype with mean recovery time of 56 seconds. By 56 days this increases to 58% with an increase in the recovery period to 85.6 seconds.

Homozygous staiB200 mutant larvae exhibit alterations in the morphology of individual axons within the segmental nerve that are not observed in wild type of heterozygous staiB200 controls. Unlike in wild type, individual axons are often bent and non-linear.

staiB200/Df(2L)Exel6015 mutant larvae exhibit alterations in the morphology of individual axons within the segmental nerve that are not observed in wild type of heterozygous staiB200 controls. Unlike in wild type, individual axons are often bent and non-linear.

staiB200/Df(2L)ED384, staiB200/Df(2L)staiL27 and staiB200 mutants exhibit large numbers of synapse retractions. stai mutants cause synapse disassembly and retraction at the neuromuscular junction and affect synapse stability at multiple muscles in the neuromuscular system.

staiB200 mutant neuromuscular junctions at muscle 4 exhibit 40% fewer boutons than in wild-type.

One copy of Khc20 increases the penetrance of the posterior penetrance "tail-flip" phenotype seen in staiB200 mutant larvae. 97% of flies exhibit a phenotype compared to 77% in the staiB200 mutants alone.

Expression of Hsap\STMN1tub.PD partially suppresses the posterior paralysis "tail-flip" phenotype seen in staiB200 mutant larvae. 40% of flies exhibit a phenotype compared to 77% in the staiB200 mutants alone. The severity is also reduced; only 9% exhibit a robust phenotype (as opposed to mild) compared to 70%.

Expression of Hsap\STMN1tub.PD rescues the altered morphology of individual axons within segmental nerve bundles of staiB200 mutant larvae.

Expression of staitub.PB partially rescues the posterior paralysis "tail-flip" phenotype seen in staiB200 mutant larvae. The onset of the bang-sensitive phenotype is delayed and less severe, with only 6% of flies exhibiting this phenotype compared to 76% in staiB200 mutants alone. The mean recovery time is reduced from ~77 seconds to ~40 seconds. Average lifespan is increase to ~69 days.

Expression of staitub.PB partially rescues the reduction in lifespan seen in staiB200/Df(2L)Exel6015 mutant larvae. Average lifespan is increased to ~65 days.

Expression of staitub.PB rescues the altered morphology of individual axons within segmental nerve bundles of staiB200 mutant larvae.

The presence of staitub.PB reduces the occurrence of observed synaptic retractions and the reduction in bouton number seen in staiB200 mutants from 70% to 10%.

Precise excision of the PBac element in PBac{5HPw[+]}stai[B200] rescues the posterior paralysis "tail-flip" phenotype, the reduced lifespan and the bang sensitive phenotype seen in staiB200 mutants.
