Description

Imprecise excision of the P{hs-hig.HA} insertion in yata25A, resulting in a 6148bp deletion of genomic sequence that removes almost the entire yata gene. A 10bp sequence of the 3' end of P-element insertion is present.

Comment:

Deletion of 6148 bp resulting from the excision of P{hs-hig.HA}yata25A. 10 bp from the 3' end of the P element remain.

Homozygous flies start to die after eclosion and show a shorter lifespan than controls.

Homozygotes show various eye defects, including an irregular arrangement of ommatidia, black coloured ommatidia and lens and bristle defects. Examination of the internal morphology of the eye reveals tissue polarity defects, abnormal rhabdomere numbers and vacuolisation at day 0 after eclosion. There is a large increase in vacuolisation in 7 day old flies raised in 12 hour light/12 hour dark conditions (vacuolisation is suppressed in constant dark conditions).

Homozygous flies show central brain volumes that are indistinguishable from wild type at day 0 after eclosion, but at day 14 after eclosion, the central brain volume of the mutant flies is significantly reduced compared to that of controls.

10% of mutant flies show a notched wing phenotype.

Expression of ApplScer\UAS.T:Ivir\HA1 or higScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.PLu partially rescues the shortened lifespan of yataKE2.1 homozygotes.

Expression of ApplScer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav.PLu rescues the reduced central brain volume seen in 14 day old yataKE2.1 homozygous adults.

yataKE2.1/yataKE2.1 Appld/Df(1)vndD38 double mutant adults show a significant reduction in central brain volume at 0 days after eclosion.

The reduction in lifespan seen in yataKE2.1 homozygous adults is more severe if the flies are also carrying Appld/Df(1)vndD38.
