Expression of ApplAβ.Scer\UAS.T:Ivir\HA1 under the control of either Scer\GAL4c739 or Scer\GAL4VT064246 (using tub-Gal80[ts] to limit the expression to adulthood) does not impair middle-term memory (measured 2hr post-training) but strongly reduces long-term memory (measured 24hr after training) tested in an aversive olfactory memory assay.
Adult specific expression of ApplAβ.Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4Mef2.247.Switch (activated in adult flies fed RU486) does not significantly affect short term memory in an olfactory aversive conditioning paradigm, compared to controls.
Adult specific expression of ApplAβ.Scer\UAS.T:Ivir\HA1 driven by Scer\GAL4Mef2.247.Switch (activated in adult flies fed RU486) significantly enhances short term memory defects in Appld/+ flies (compared to Appld/+ flies or to controls not fed RU486); there are no significant effects on learning, olfactory acuity or shock reactivity.
30 day old flies expressing ApplAβ.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4GMR.PF have aggregates in the retina (these aggregates are not seen in wild-type controls). These aggregates have characteristics of amyloid deposits. These 30 day old flies also show extensive vacuolisation of the retina.
Expression of ApplAβ.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4Appl.G1a results in the presence of toluidine blue-positive deposits, vacuoles and dark shrunken nuclei in the brains of 30 day old flies. No gross abnormalities are seen in these flies at 2 days after eclosion.
6 day old flies expressing ApplAβ.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4elav-C155 have a significantly lower performance index in a fast phototaxis assay than control flies and the performance continues to decline with age in the mutant flies.

Adult flies expressing both Nep1KK101227 and ApplAβ.Scer\UAS.T:Ivir\HA1 under the control of Scer\GAL4c739 (combined with tub-Gal80[ts] to limit expression to adulthood) do not show any defects in middle-term memory (MTM) tested in an aversive olfactory memory assay 2hr post-training, whereas with the Scer\GAL4VT064246 driver, the MTM is significantly reduced.
