Expressing GckIIIKK101915 under the control of Scer\GAL4C587 (together with Dicer-2, for a more efficient RNAi) does not induce any obvious L3 spermatogenesis defects.

Third instar larvae expressing GckIIIKK101915 under the control of Scer\GAL4btl.PS exhibit a tracheal morphogenesis phenotype, with the terminal cells displaying a transition zone tube dilation defect. Ectopic septate junctions are seen in these terminal cells; the frequency with which septate junctions extend into the terminal cell transition zone is increased compared to wild type, with junctions extending into between 32% (vari staining) and 42% (Fas3 or cora staining) of cells compared to only 4% in wild type. Additional terminal cells display punctate septate junction staining and short discontinuous patches of septate junction staining can be seen 10s or 100s of microns away from the transition zone.

There is a significant reduction in chromosome instability tolerance induced by mad2GD16427 expression, when GckIIIKK101915 is expressed in imaginal wing discs under the control of Scer\GAL4en.PU or Scer\GAL4hh.PU.

Heterozygosity for Df(3R)Exel6174 enhances the penetrance of the transition zone tube dilation defect seen in third instar larval trachea expressing GckIIIKK101915 under the control of Scer\GAL4btl.PS.

Heterozygosity for Ccm3ins enhances the penetrance of the transition zone tube dilation defect seen in third instar larval trachea expressing GckIIIKK101915 under the control of Scer\GAL4btl.PS.

Heterozygosity for Ccm3rev does not enhance the penetrance of the transition zone tube dilation defect seen in third instar larval trachea expressing GckIIIKK101915 under the control of Scer\GAL4btl.PS.
