The expression of PdhbKK107865 under the control of Scer\GAL4GMR.PU induces a rough eye phenotype.

Expression of PdhbKK107865 under the control of Scer\GAL4GMR.PS results in rough eye phenotype associated with interommatidial bristle loss and ommatidium fusion in adult flies, the phenotype is made more severe with two copies of PdhbKK107865. No increase in the level of apoptosis (visualized by [*]Casp3 staining) is observed in third instar larval eye discs upon PdhbKK107865 expression.

Locomotor activity is perturbed by Scer\GAL4elav.PLu-driven expression of PdhbKK107865 both at the larval stage (the average crawling speed is slightly but significantly reduced and the larvae frequently show aberrant crawling behavior with abnormal path patterns) and the adult stage where the climbing ability of the flies is significantly reduced already at 5 days post-eclosion and rapidly declines further with age. The lifespan of PdhbKK107865-expressing adult flies is also significantly shorter compared to controls.

Third instar larvae expressing PdhbKK107865 under Scer\GAL4elav.PLu also display neuromuscular junction (NMJ) morphological defects: the NMJ branch length along with the number of Ib boutons is reduced, but the Ib bouton size is increased. The knock-down larvae also exhibit defects in photoreceptor axon innervation of the lamina and medulla and increased mitochondrial fragmentation in the brain. The abnormal fragmented and punctiform morphology of brain mitochondria is also seen in adult flies expressing PdhbKK107865 under Scer\GAL4elav.PLu

Number of midgut cells per MARCM clone expressing PdhbKK107865 under the control of Scer\GAL4Tub.PU is similar to control adults under calorie-restricted condition.
