UASt regulatory sequences drive expression of an inverted repeat.
The expression of TfAP-2GD4684 under the control of Scer\GAL4elav.PU, but not of Scer\GAL4tim.PU, Scer\GAL4Pdf.PU, Scer\GAL4Gad1.PU, Scer\GAL4104Y Scer\GAL4GMR23E10, Scer\GAL4GMR21B10, Scer\GAL4GMR20D06, Scer\GAL4Tdc1.PU, Scer\GAL4cv-c-C5, Scer\GAL4GMR20F11 or Scer\GAL4Gad1.PU, leads to a significant decrease in sleep time, which is explained by a significant decrease in night-time sleep, as compared to controls; their adult brains exhibit slimmer or disrupted mushroom body alpha/beta lobe axonal projections, as compared to controls. Expression under the combined control of Scer\GAL4elav.PU and Dicer-2 (for a more efficient RNAi) leads to a more severe sleep phenotype, almost abolishing the night-time sleep. Adulthood-only expression under the combined control of control of Scer\GAL4elav.PU and Gal80[ts] (for temporal regulation of expression) also leads to a significant decrease in sleep time, which is more severe by the additional expression of Dicer-2 (and more efficient RNAi).
