Description

Imprecise excision of the progenitor insertion, resulting in a 4717bp deletion (coordinates 2L: 20792456...20797173) that removes part of the vari transcription unit. 416bp of the original insertions remains. The deletion also removes the 3'UTR and the 3 C-terminal amino acids of Pomp.

Comment:

4717bp deletion resulting from the imprecise excision of P{EP}variGE13049. Removes most of vari coding sequence and the last three amino acids of Pomp. Position of deletion on reference sequence inferred by FlyBase curator.

No significant increase in defasciculation defects in intersegmental nerve b motor axons is observed in vari48EP heterozygous embryos compared to controls.

vari327/vari48EP transheterozygous embryos display increased frequency of defasciculation defects in intersegmental nerve b motor axons.

The stage 15 embryonic hindguts of vari48EP homozygotes are of normal length but present a lower angle of curvature, as compared to controls.

Late embryonic lethal.

Homozygous embryos show loss of integrity of the transepithelial barrier; dextran dye is detected within the tracheal lumen of mutant embryos within 30 minutes of injection. The lumens of stage 16 trachea can be significantly dilated in mutant embryos compared to wild type, with large dilations being present along the dorsal trunk, but the structure of the lumenal cuticle and taenideal ridges is unaffected. Some lateral branches appear disconnected from the dorsal trunk.

The ultrastructure of the lateral membranes and septate junctions of epithelial cells in the hindgut is similar in mutant and wild-type embryos.

vari48EP/vari03953b embryos have large dilations along the dorsal trunk.

Homozygous clones are not recovered in the adult eye, although the eyes contain fused ommatidia, missing bristles and duplicated bristles.
