Expression of RhoGAP18BRC.Scer\UAS under the control of Scer\GAL4whir3 has no effect.
Adult overexpression of RhoGAP18BRC.Scer\UAS in wild-type flies (Scer\GAL4whir3/+, Scer\GAL80ts.αTub84B; Avic\GFPScer\UAS) does not change ethanol-induced hyperactivity.

Expression of RhoGAP18BRC.Scer\UAS under the control of Scer\GAL4whir3 in RhoGAP18B3 mutant males results in nearly normal ethanol sedation. This behavioural rescue is manifested as a reduction in the time required for 50% of the flies to reach sedation. In addition, RhoGAP18BRC.Scer\UAS expression also increases the viability of the RhoGAP18B3 mutant from 31% to 83%.
Expression of RhoGAP18BRC.Scer\UAS under the control of Scer\GAL4whir1 in RhoGAP18B1 mutants ameliorates the ethanol-resistance phenotype of RhoGAP18B1. The phenotype rescue is, however, incomplete, probably due to the fact that expression of RhoGAP18BRC.Scer\UAS under the control of Scer\GAL4whir1 is low.
Expression of RhoGAP18BRC.Scer\UAS under the control of Scer\GAL4whir3, using Scer\GAL80ts.αTub84B to block expression of Scer\GAL4whir3 at the permissive temperature (16[o]C), except for three days in adulthood, where the restrictive (25[o]C or 29[o]C) temperature is used to allow expression of Scer\GAL4whir3 to drive RhoGAP18BRC.Scer\UAS reveals a complete rescue of the RhoGAP18B3 ethanol-induced sedation phenotype. The reciprocal experiment, where the flies are raised at 25[o]C and then placed at 16[o]C for 3 days prior to behavioural testing fails to rescue the RhoGAP18B3 ethanol-induced sedation phenotype and in fact worsens the phenotype compared to control flies.
